You just need to indicate the tense (tempo verbal) once. So, in the case, you already have the auxiliar "Does" indicating that this sentence is in the 3rd person; therefore, you don't need to indicate this again in the main verb - the auxiliary already did this.
So, the correct would be: "Does she want to come?"
